en.Wedoany.com Reported - On August 24, Johnson & Johnson announced that the U.S. Food and Drug Administration (FDA) has approved IMAAVY® (nipocalimab-aahu) for the treatment of warm autoimmune hemolytic anemia (wAIHA) in adults and pediatric patients aged 12 years and older who have received or are currently receiving glucocorticoid therapy. This approval makes IMAAVY the world's first therapeutic drug specifically approved by the FDA for this disease.

Warm autoimmune hemolytic anemia is a rare and potentially life-threatening autoantibody-mediated hematologic disease. In affected patients, immunoglobulin G (IgG) autoantibodies mistakenly attack and destroy red blood cells, leading to severe anemia, extreme fatigue, and a high likelihood of disease relapse. Previously, no targeted therapy had been specifically approved for wAIHA in the United States, and clinical management primarily relied on off-label treatments such as glucocorticoids, rituximab, and immunosuppressants, with some patients showing inadequate responses.

IMAAVY is a selective monoclonal antibody that blocks the neonatal Fc receptor (FcRn). Its mechanism of action involves blocking FcRn-mediated recycling of IgG antibodies, thereby accelerating the clearance of pathogenic autoantibodies while preserving baseline B-cell function. This mechanism differs from traditional B-cell-depleting agents and offers a more targeted therapeutic strategy.

This approval is based on results from the pivotal Phase 2/3 ENERGY clinical trial. Data showed that a higher proportion of patients treated with IMAAVY achieved a durable hemoglobin response compared to the placebo group; a significant increase in hemoglobin levels was observed as early as Week 1 of treatment, with a mean increase of 1 g/dL, and improvements in patient fatigue scores (FACIT-Fatigue) were also noted at Week 24. The overall safety profile was manageable.

The product had previously received FDA Priority Review and Orphan Drug Designation. IMAAVY was already approved in the United States for generalized myasthenia gravis (gMG), and this expansion into the wAIHA indication further broadens the application of this FcRn inhibitor in the field of autoantibody-mediated rare diseases. The drug has not yet been submitted for marketing authorization in China.
